The priests needed to cover 100% belief in the city you have to do with the city's population to have the temple. So for example if you have town hall level 20 and your citizens is 1878 then you will need 1878 / 5 = 375.6 priests. Of course if we put 375 priests will not cover 100% because we still need 0.6 priests. So should we rounded up so we will need 376 priests.

Can I use the miracle and then just demolish the temple.
This is done but assuming you have a city with an empty space and just use it to build a temple where you need a miracle has no substance. So if you have a city level of 15 will need to upload the temple so that you can cover 1,200 citizens, which means that you will need 240 priests. Until then it will throw the miracle the other residents of your island. Besides this will take time to be able to cover all 5 levels of the miracle. This is possible and the reason for this is because a new colony will need a small temple and a city hall is 1 level with the original citizens 60 (to 130 according to the research you do the cities you have and the corruption the people vary) so you will need 12 priests to cover 100% faith in your town. So you will need as much time as you want to load the goods to your new city. Sure you need wood and glass. In time you want to get to the island of your choice and the time required the church to level 1 (40 minutes). Of course with that will build the temple will begin to fall the miracle directly to the island as you see in the picture.Of course this does not affect much the other players as your new city will be 1 level.
Like I said before, depending on Population and corruption that you have perhaps you need and another level of the temple to cover people means that you will need another 40 minutes to the next level. In my case with 8 cities and mobile city (9th) for the miracle it took me 3 levels of the temple to be able to cover 100% faith of the population of the town which means a time of 2 hours (120 minutes) approx. You will also need some time before you can obtain the faith necessary to activate the miracle.

Total you need 3 hours + the time it takes to "fill" the miracle to build a colony to take advantage. (Depends the cities and research you've done)
If you turn the miracle of mobile colony and then leave the colony what?
In this case there is nothing happend the miracle continues to operate normally.
The reuse of the miracle reduced if I have the same temple wonder on the same island?
Yes reuse time is reduced if you have 2 temples in the same miracle. As many cities with temples you have in the same miracle the more reduced the time it will need to use it again.
The time is reduced if I have in the same miracle a temple, but to a different island?
Yes time is reduced even if you have a temple in the same miracle, but to to another island. Just need to watch this. The time will be reduced accordingly levels of 2 miracles. For example if you have a miracle at 5 and another miracle on the level 2 Poseidon then reuse falls on 17 hours and 8 minutes. If the miracle move from level 2 to level 3, the time will be reduced to 15 hours. To you have as fast as possible reuse should have the same levels of the miracle on the islands.
If I have less faith can I use the miracle?
Yes you can turn the miracle will just get smaller levels of miracle. For example, if cover 20% of faith in your city then you can turn the miracle at the level 1 for the 40% level 2 for the 60% level 3 for the 80% level 4 and 98% over the level 5.
